Definitely the best method would be to use immunostaining, this is a protocol that makes sure the use of Triton to break down cell      
membranes. Immunostaining can be used to see which cells are positive for dynein. It also helps you to make sure to counterstain to make cells vissible use hematox when you use light microscopy and DAPI for fluorescence microscopy.
