There are 3 stages in translation/protein synthesis:

The initiation stage during which the mRNA binds to the small ribosomal subunits and the tRNA carrying amino acid binds to the mRNA at the start codon. Lastly, the large subunit of the ribosome binds to the structure to complete the initiation stage.The elongation involves the movement of the complex across each codon of the mRNA, the release of their corresponding amino acids by the tRNA. and the formation of a peptide bond between the amino acids.The termination stage during which there exists no corresponding amino acid for the codon. The system reads such a codon as a sign to stop the translation process.

Mupirocin works by inhibiting one of the tRNAs. Specifically, it inhibits the tRNA carrying an amino acid known as isoleucine. Thus the translation process is disrupted and the protein synthesis truncated. In other words, mupirocin works by cutting short the elongation process of translation.
