Move the filled cuvette containing the 100 µM solution to the spectrophotometer and load into the sample chamber as before. With the spectrophotometer in the Full Spectrum Mode, obtain a spectrum of the EDMAB solution, and click the REC button to display it below. You might need to scroll this text area to see the chart. Refer to the note you made in your lab notebook about the wavelength of maximum absorption for EDMAB in ethanol. Pay particular attention to the sharp peak around 300 nm. Notice how much stronger this peak is than anywhere else in the range 200-600 nm. You need to locate this peak experimentally by adjusting the wavelength setting of the spectrometer. Switch the instrument back to Single Wavelength Mode. Find the absorbance maximum for the sample by entering wavelengths and reading absorbancies until you have found the maximum. There may be more than one maximum, so explore the frequency range to be certain you have found the wavelength with overall maximum absorbance. You should be able to determine the maximum value to within 1 nm. The strongest absorption peak is quite sharp, and you will need to explore carefully to find the wavelength which has the strongest absorption. This process need only be performed now at the beginning of the analysis. Hint: The correct wavelength will be slightly greater than 300 nm, and the absorbance of the sample at the maximum will be greater than 2 a. u.
